Genetic Analysis and Quality Effects of 1Dx5 Subunit Silent in Transgenic Wheat

High-molecular-weight glutenin subunit(HMW-GS) variation is related with the wheat processing quality.To investigate the genetic behavior of silent 1Dx5 gene in RNAi transgenic wheat, the transgenic wheat line Glu-lDx5-RNAi was used to hybridize with two soft wheat varieties Yang-mai 18 and Yangmai 13 which were adopted as recurrent parents in backcrosses.The HMW-GS distribution in parents,and their F_1,F_2 and BC_1F_1 progenies were analyzed by the SDS-PAGE technique. Results showed the silent effect of 1Dx5 gene is dominant and can stably inherit in transgenetic progenies. The segregation ratio of lines without 1Dx5 subunit and lines with 1Dx5 subunit is 13:3 and 3: 1 in F_2 and BC_1F_1 progeny,respectively.The silent 1Dx5 gene by RNAi may inherit as a single dominant genetic factor.Quality tests of transgenic wheat varieties and receptor Bobwhite showed that the total protein content of transgenic lines was 13.28%,which was slightly higher than that of the receptor lines with protein content of 12.83%.Moreover,the hardness index and SDS sedimentation value reduced by 3.13 and 3.7 mL in transgenic wheat varieties than that in receptor,respectively.
