Expression changes of mmp-2,9 and timp-1 in myocardial with pulmonary hypertension

[Objective]To explore the role of the matrix metalloproteinases(MMPs)and tissue inhibitor of metalloproteinase(TIMPs)in the left and right ventricle expression and remodeling on rats with Pulmonary hypertension.[Methods]60 SD rats were randomly divided into 5groups.12 SD rats were in each group.There were one control group and 4 experimental pulmonary hypertension(PAH)groups.We took samples at 4 time Points(MCT were injected at the 1,2,3 and 4 week).MCT would be injected at 1,2,3 and 4 week after experimental pulmonary hypertension(PAH),the corresponding experimental groups were regarded as PAH1,PAH2,PAH3 and PAH4 experimental groups.The activity of MMPs was measured by zymography.The protein and mRNA of MMP-2,-9 and TIMP-1 were determined by Western-Blot an d RT-PCR.[Results](1)MMP-2 and 9 mRNA reached peak at 1 week after Pulmonary hypertension,and gradually declined to 4 weeks,but compared with the control level.The experimental group was still higher,there were significant differences significance(P﹤0.01).TIMP-1 mRNA reduced in the experimental group from the first to the fourth week.Compared with the control level,there were significant differences significance(P﹤0.01).(2)In the left ventricle,MMP-2 and 9 Protein activity slightly increased in rat Pulmonary hypertension,the expression slightly increased,while TIMP-1 Protein content was also slightly reduced.But there was no significantly difference with the control group,and they could not lead to left ventricular remodeling.[Conclusion]MMP-2,9 Protein activity and its expression increased in rat right ventricle with Pulmonary hypertension.But TIMP-1 Protein activity and expression decreased in rat right ventricle with Pulmonary hypertension.They may be related to right myocardial remodeling.Not left ventricular remodeling.
