Prediction of B- and T-cell epitopes of gB gene of duck plague virus and prokaryotic expression of its major antigen determinants.

B-and T-cell epitopes of duck plague virus(DPV) gB gene(GenBank accession No.EF608147) were predicted by bioinformatics softwares.A fragment encoding gB major epitopes was amplified by PCR and subcloned into the prokaryotic expression vector pET-28a(+).The constructed recombinant was transformed into Escherichia coli BL21(DE3) competent cells and induced to express.The results showed that the recombinant protein with molecular weight of 46ku was expressed in inclusion bodies and accounted for 30% of the total proteins.Western-blot analysis showed that the expressed protein could be recognized specifically by polyclonal rabbit anti-DPV serum,indicating that the expressed protein possessed strong reactinogenicity.
