The Effect of Lysophosphatidie Acid on the Gene Expression and Activityof Matrix Metalloproteinases-9 in THP-1 Cell Line

Objective To study the effect of lysophosphatidie acid(LPA)on the gene expression and activity of matrix metalloproteinases-9(MMP-9)and the activity of NF-κB p65 protein in inTHP-1cell line and to investigate the role of MMP-9 invovled in the atherosclerosis caused by LPA.Methods Human THP-1cells were stimulated with LPA at different concentrations(0~10 μmol/L),the gene expression and activity of MMP-9was measured by RT-PCR and gelatin zymography respectively,the activity of NF-κB p65 was detected by westernblot according to the expression level of NF-κB p65 protein in cell nucleus.Results LPA upregulated the gene expression and activity of MMP-9,activated the NF-κB p65 in a dose-dependent manner,the peak point occurred at 1 μmol/L LPA and decreased at higher LPA concentrations.Conclusion LPA may increase the expression and activity of MMP-9 at transcription level involving activation of NF-κB pathway in THP-1cells which may contribute to the atherosclerosis process.
