Measurement of Rest:Activity Rhythms with and without MPEP Treatment Flies were entrained on standard cornmeal, agar-molasses-yeast medium at 25°C. For activity assays, male flies were entrained to a 12:12 hr light:dark cycle for a minimum of 5 days prior to loading in glass tubes for activity monitoring using a Trikinetics (Waltham, MA) system interfaced with an Apple computer. Data were subsequently analyzed using the Clocklab (Actimetrics, Evanston, IL) software package. Rhythmicity was determined by visual inspection of the actograms and periodograms and FFT analysis. To test the ability of MPEP to rescue rest:activity rhythms in dfmr1 and FS flies, mutant and control flies were fed MPEP during development and/or as adults and/or in the food contained within the glass tubes used in the activity monitors. Concentrations of MPEP used ranged from 1 to 200 μM.
