A majorfocusofstudies thatcenter on regeneration oftheperiodontium istodetermine the efficacy oftheuseofpolypeptide growth factors. Platelet- derived growth factor hasbeenreported tobeapossible agentforclinical use.PDGFhasvarious isoforms. Therefore, wedecided tostudythemitogenic and chemotactic responses ofhumanperiodontal ligament (PDL) cells torecombinant humanPDGF-AB, AA,andBB. Addition ofeachisoform ofPDGFtoinvitro mitogenesis assays induced PDLcell proliferation inadose-dependent manner. Themaximummitogenic effect wasevident atthe concentration of100ng/mL. Inthese assays, PDGF-BB was found tobethemostpotent mitogen. PDGF-ABelicited an intermediate response, andPDGF-AAwastheleast effective. Theresults ofchemotaxis assays closely parallel those ofthemitogenesis assays. PDGF-BBexhibited the mostpotent chemotactic effect. Themaximal effect was observed at10ng/mL. Thefindings ofthese experiments indicate that PDGF-BBismoreeffective thantheother isoforms inpromoting mitogenesis andchemotaxis ofPDL cells invitro, andmaytherefore beasuitable ethical pharmaceutical foruseinperiodontal regeneration procedures.
