Effects of CXCR4 gene silencing by small interfering RNA on invasiveness of bladder cancer T24 cells

Aim:To investigate the effects of CXCR4 gene silencing by small interfering RNA on invasiveness of bladder cancer T24 cells.Methods:Using LipofectamineTM2000,CXCR4 siRNA was transfected into T24 cells in vitro(siRNA transfect group). Blank control group and negative control group were established.RT-PCR was used to detect the expression of CXCR4 mRNA in T24 cells. Westerm blot was used to detect the expression of CXCR4 protein. Boyden chamber experiment was used to count the membrane-permeable cells.Results:The levels of CXCR4 mRNA and protein expression,and the number of membrane-permeable cells among the 3 groups had significant differences(F=140.822,473.079,133.247,P<0.001),which were lower in siRNA transfect group than those in the control groups, and the invasive ability of the cells in siRNA transfect group decreased(P>0.05).Conclusion:Using siRNA to down-regulate CXCR4 gene can reduce the invasive and proliferative power of T24 cell line.
