HPLC determination of three effective ingredients in Corydalis decumbens(Thunb.)Pers.injection

Objective:To accurately quantify three effective ingredients(protopine,tetrahydropalmatine,and palma- tine)in Corydalis decumbens(Thunb.)Pers.injection by HPLC.Methods:HPLC analysis was performed on a Dia- monsil C_(18)reverse-column(5 μm,250 mm×4.6 mm)with a gradient of the following solvent mixtures:solvent A, acetonitrile;solvent B,acetic acid-triethylamine(dissolve 30 mL acetic acid in 1000 mL water,add 8 mL triethyl- amine).The flow rate was 1 mL·min~(-1),the deteetion wavelength was 280 nm,and the column temperature was 25 ℃.Results:The calibration curves of protopine,tetrahydropalmatine,and palmatine were linear in the ranges of 8.56-171.2 μg·mL~(-1)(r=0.9998),7.12-142.4 μg·mL~(-1)(r=0.9997),6.72-134.4 μg·mL~(-1)(r= 0.9996),respectively.Recoveries were 99.3%-100.4%(RSD≤1.1%),99.8%-100.0%(RSD≤1.5%), 99.4%-99.9%(RSD≤0.9%),respectively.Conclusion:The method is accurate,rapid,and can be used for de- termination of protopine,tetrahydropalmatine,and palmatine in Corydalis decumbens(Thunb.)Pers.injection.
