The relationship between the expression of ERCC1 genes and Fe_3O_4-magnetic nanoparticals with drug resistance in ovarian carcinoma cells

Purpose To determine whether Fe3O4-MNPs can reverse the cisplatin(DDP) resistance to ovarian carcinoma cells(SKOV3/DDP)and to explore its mechanisms.Methods The SKOV3/DDP strains were divided into DDP treatment(DDP group),Fe3O4-magnetic Nanoparticals treatment(Fe3O4-MNPs group),and DDP+Fe3O4-MNPs treatment(DDP+Fe3O4-MNPs group)and control group.The apoptotic ratios were detected by flows cytometry(FCM),and the intracellular platinum levels in different groups were detected by ICP and the expressions of excision repair cross complement-1(ERCC1) were detected by reverse transcription polymerase chain reaction(RT-PCR) in different groups.Results In the DDP+Fe3O4-MNPs groups,the apoptotic ratios and the intracellular platinum levels were higher.The expression of ERCC1 was decreased compaired with other groups.Conclusion Fe3O4-MNPs can reverse the DDP resistance to the ovarian carcinoma cells(SKOV3/DDP),and the effect may be ascribed to increase the intracellular platinum levels and decrease the expressions of ERCC1 gene.
