In 2001, with other members of the Pharmacology group of the Department of Experimental Medicine (University of Perugia), Ursula Grohmann discovered a crucial immunosuppressive role in tumor immunity for the enzyme indoleamine 2,3-dioxygenase (IDO) expressed by dendritic cells. Since then, she continued to study many aspects of IDO’s biology not only in experimental tumor systems, but also in autoimmune diseases, allergy, and chronic inflammation. Grohmann's group was the first to describe several novel aspects of IDO’s biology, most of which were presented in several publications of excellence at the international level. In particular, Ursula Grohmann's group has greatly contributed to the subject of immune tolerance, providing evidence that (a) tryptophan catabolism in tolerogenic dendritic cells can exert profound immunoregulatory effects on T lymphocytes as well as on otherwise immunostimulatory dendritic cells (DCs); (b) tryptophan catabolism is defective in autoimmune/inflammatory diseases; (c) kynurenines, the products of tryptophan catabolism, can be used therapeutically in chronic inflammation; (d) tryptophan catabolism represents a fundamental mode of action of specific immunosuppressive drugs, including glucocorticoids; (e) indoleamine 2,3-dioxygenase is not just a tryptophan catabolizing enzyme but also a signaling protein fundamental in mechanisms of long-term immune tolerance. Grohmann's group, together with Paolo Puccetti, currently constitutes one of the top world leaders in research related to IDO.
