Dr. Nikolovska-Coleska’s lab focuses on chemical genomics, discovery and application of active chemical compounds for the interrogation of biological systems, and developing molecularly targeted therapeutics for the treatment of human cancer. Towards discovery and development of small molecule inhibitors targeting protein-protein interactions, Dr. Nikolovska-Coleska applies different strategies for identification of hit and lead compounds, including high throughput screening, virtual screening and structure-based design. For this purpose series of complementary biochemical, biophysical, functional and cell based assays are developed and used for screening of compound libraries, as well as for validating and characterizing of identified small molecules. Such well-characterized small-molecules will serve as (a) pharmacological tools for elucidation and understanding of the functions of corresponding targets and (b) promising lead compounds for further optimization toward our ultimate goal of developing new anticancer drugs using interdisciplinary approach combining structural biology, medicinal chemistry and functional biology. Dr. Nikolovska-Coleska research is primarily focused on understanding the protein-protein interactions involved in controlling the programmed cell death (Bcl-2 family of proteins) and regulating epigenetic modifications (histone methylation through histone lysine methyltransferases).
