I have diversified and successful research experiences in immunology, virology and cancer studies. Currently I am working on the development of novel immunotherapies for cancer treatment based on leukocytes signaling modulation.
Intriguingly, SLFN11 was also found to be the only gene whose expression significantly sensitizes cancer cells to apoptosis upon treatment with DNA-damaging agents in the form of chemo-/radio- therapies. The expression of SLFN11 was silenced or significantly down-regulated in about half of all primary tumors regardless of their types, leading to their resistance to chemo-/radio- therapies. My work discovered that human SLFN11 sensitizes cells to apoptosis upon DNA damages through the cleavage of type II tRNAs including all serine and leucine tRNAs. One of these tRNAs in particular, “tRNA-Leu-TAA,” is of extremely low abundance in the cell, but nevertheless used frequently in the protein syntheses of most genes responsible for DNA damage repairs, rendering those genes highly susceptible to the inhibition by SLFN11. Thus, silencing of SLFN11 expression in cancer cells significantly enhanced the repair of DNA damages and the survival of those cancer cells upon chemo-/radio- therapies. Targeting the drug resistance caused by SLFN11 deficiency in cancer cells offers a novel approach of synthetic lethality for cancer treatment.
